چکیده

Immunotherapy may provide valid alternative therapy for patients with hormone-refractory metastatic prostate cancer. However, if the tumor environment exerts a suppressive action on antigen-specific tumor-infiltrating lymphocytes (TIL), immunotherapy will achieve little, if any, success. In this study, we analyzed the modulation of TIL responses by the tumor environment using collagen gel matrix-supported organ cultures of human prostate carcinomas. Our results indicate that human prostatic adenocarcinomas are infiltrated by terminally differentiated cytotoxic T lymphocytes that are, however, in an unresponsive status. We demonstrate the presence of high levels of nitrotyrosines in prostatic TIL, suggesting a local production of peroxynitrites. By inhibiting the activity of arginase and nitric oxide synthase, key enzymes of L-arginine metabolism that are highly expressed in malignant but not in normal prostates, reduced tyrosine nitration and restoration of TIL responsiveness to tumor were achieved. The metabolic control exerted by the tumor on TIL function was confirmed in a transgenic mouse prostate model, which exhibits similarities with human prostate cancer. These results identify a novel and dominant mechanism by which cancers induce immunosuppression in situ and suggest novel strategies for tumor immunotherapy.

Immunohistochemical evaluation of lymphocyte types infiltrate into the canine seminomas

BACKGROUNDS: Seminoma is frequently observed in human and canine testes especially in cryptorchids. Canine and human seminomas are typically associated with leukocytic infiltration. OBJECTIVES: We aimed to identify the type of lymphocytes that infiltrate to seminomas. METHODS: Tumor infiltrating lymphocytes were evaluated by immunohistological techniques in 5 dogs with diffuse seminoma. Routine...

CD40L confers helper functions to human intra-melanoma class-I-restricted CD4+CD8+ double positive T cells

Although CD4+CD8+ double positive (DP) T cells represent a small fraction of peripheral T lymphocytes in healthy human donors, their frequency is often increased under pathological conditions (in blood and targeted tissues). In solid cancers such as melanoma, we previously demonstrated an enrichment of tumor reactive CD4lowCD8highαβ DP T cells among tumor-infiltrating lymphocytes of unknown fun...

Potential Role for T Cells in Angiogenesis A Human Cancers Express Vascular Endothelial Growth Factor: Peripheral Blood T Lymphocytes and Lymphocytes Infiltrating

CD3* peripheral blood T lymphocytes were evaluated for expression of vascular endothelial growth factor (VEGF), an endothelial cell mitogen and potent angiogenic factor. VEGF mRNA expression was confirmed in CD3+ cells and Jurkat cells, a human T-cell line, by reverse transcriptionPCR and in ( ÕU and CDS' T cell subtypes by Northern blot hybridiza tion. Steady-state levels of VEGF mRNA were in...

Smad and NFAT pathways cooperate to induce CD103 expression in human CD8 T lymphocytes.

The interaction of integrin αE(CD103)β7, often expressed on tumor-infiltrating T lymphocytes, with its cognate ligand, the epithelial cell marker E-cadherin on tumor cells, plays a major role in antitumor CTL responses. CD103 is induced on CD8 T cells upon TCR engagement and exposure to TGF-β1, abundant within the tumor microenvironment. However, the transcriptional mechanisms underlying the co...

A Novel Antagonist of the Immune Checkpoint Protein Adenosine A2a Receptor Restores Tumor-Infiltrating Lymphocyte Activity in the Context of the Tumor Microenvironment

BACKGROUND Therapeutic strategies targeting immune checkpoint proteins have led to significant responses in patients with various tumor types. The success of these studies has led to the development of various antibodies/inhibitors for the different checkpoint proteins involved in immune evasion of the tumor. Adenosine present in high concentrations in the tumor microenvironment activates the i...
